ABSTRACT:
The Sohl lab explores the molecular consequences of enzyme mutations implicated in cancer, using enzyme kinetics, X-ray crystallography, and cellular models to establish the mechanisms of dysfunction. One focus is on the role of isocitrate dehydrogenase 1 mutations in lower grade gliomas. We find that the catalytic features of these mutations dictate key tumor phenotypes, and we are currently using lipidomic and metabolomic methods to illuminate new downstream consequences in glioma models.
